Cas9 lentiviral expression vectors can be used to efficiently transduce mammalian cell lines to engineer gene knockout stable cell lines. The Cas9 endonuclease, when programmed with a single gRNA, leads to a DNA double-strand break (DSB) at a desired genomic location.  The cell then activates endogenous DNA repair processes, either non-homologous end joining (NHEJ) or homology-directed repair (HDR), to mend the targeted DSB. Choose the optimal promoter, selection marker, and fluorescent reporter for your CRISPR knockout experiment.
